This "ARRA Supplement" to K08DK76845 "Growth Regulatory Mechanisms of KLF in Barrett's Epithelium" is a request to expand the personnel support for the aim 3 of K08 to determine the in vivo relevance of KLF11 on the neoplastic transformation in Barrett's mucosa. In the last 2 years, intramural (non-federal) funds helped support the extra effort of our senior research technician beyond what was required by K08. This helped us stay ahead of our K08 time line to essentially complete the aim 1 and 2 of K08. Due to the finicial crisis at our institute, the intramural support is declining and we are struggling to maintain this momentum. We are concerned that we may not be able to sustain the gains made in the past 2 years, because the aim 3 is a labor intensive in-vivo aim. To overcome this we are requesting an expansion of personnel support using this administrative supplement, which will help cover the effort of our research technician. This will allow a rapid expansion and characterization of the KLF11 -/- mice colony as well as optimization of microsurgical intervention, thus increasing the pace of development of preneoplastic lesions, which is the central goal of aim 3. Our lab's current and future goals as well as the scientific community will certainly benefit from this optimized in-vivo model. The scope of the peer-reviewed K08 will not change because the genetically modified animals will undergo the same surgical procedure as outline in the K08 but the increased pace of experiments will allow us approximately 6 to 12 additional months to generate the preliminary data for our future R01. In summary, this supplement application fits in the scope of the parent grant. This will ensure that the goals of the parent grant will be accomplished within the proposed timeframe and allow us to maintain the progress of the parent grant that we achieved in the first two years. Our research team members are among few labs that have this microsurgical experience to accelerate the temp of scientific research. This clearly aligns with the goal of successful transition from K08 to an independent R01, which fit with the priorities of NIDDK. Finally this support will help with job retention, which is the mandate of the Recovery Act.
