Lung involvement is currently the leading cause of morbidity and mortality in scleroderma (SSc) patients. Interstitial lung disease (ILD) is the most common pulmonary manifestation and may result from an abnormal immune response carried out by pro-fibrotic effector T cells. In this study, entitled ?T cell response in scleroderma lung disease: from pathogenesis to clinical care?, we will prospectively study the features and the magnitude of the immune response, and in particular of the pro-fibrotic T cell subsets, in SSc patients before and during the progression to pulmonary fibrosis.

The FFR was signed on 10/24/11. The primary objective of this administrative supplement was to provide support for a full time research technician assisting the principal investigator (Dr. Boin) in the organization and conduction of the experimental portion of the parent grant (K23 AR055667). The new position has been crucial in order to start enrolling patients immediately from the beginning of the supported period. His contribution has been essential to maintain a steady patient recruitment rate and to organize on a daily base the storage of samples and the filing of generated data. The technician has gained independence in term of acquiring data with the flow cytometer, and became involved in the analysis of available data using FlowJo, a cutting edge flow cytometry analysis software. During the last quarter of this grant, the project has been progressing swiftly. At this time we have enrolled 140 patients in visit #1, 100 in visit #2, 60 in visit #3, 40 visit #4 and we are starting now with visit #5 (interval between visits is 6 months). The collection of data as well as the organization of stored samples collected at each visit is ongoing. We generated a bank of different specimens such as DNA/RNA extracted from peripheral blood mononuclear cells (PBMCs), the supernatant from stimulated PBMC for cytokine secretion profiling, the serum and plasma for each patient, at each visit, which will be used for nested studies. The research technician supported through this ARRA funding will remain the crucial quarterback of all the streamlined procedures for sample collection, analysis and storage even after the grant has expired. This project has ended and all funds were fully exhausted. |
