This project is in association with the parent grant to launch the Schizophrenia Research Center at Johns Hopkins, in response to PAR-07-434 Silvio O. Conte Center to develop collaborative research in neuroscience of mental disorders (P20). The overall goal of this center grant is to provide a multifaceted approach towards understanding the molecular pathology of schizophrenia. Productive links from basic molecular neuroscience to behavioral neuroscience, genetics, and clinical psychiatry cannot be achieved by individual RO1 studies. Thus, in this P20 center, we stated that an animal behavioral core and a human genetics core to be the key structures in the original proposal. Since the center was started in 2008, three projects have been developed very efficiently and we have made significant progress, according to the proposed plans. This project is to keep a tempo in validating novel biological hypotheses and targets identified in these three projects in animal models and human genetic approach according to the rapid progress in the Projects. Through this project, we hope to obtain important biomarkers for the disease and targets for possible therapeutic strategies toward the disease.

With the support of the ARRA supplementary fund, we have reinforced genetic core in which we have selected genes towards identification of rare genetic variants associated with schizophrenia by direct sequencing. We have selected RPGRIP1L and DISC1 for the intensive sequencing. This selection is also supported by many biological assays that show relevance of the genes in the pathophysiology of schizophrenia. We now complete the first analysis. In parallel, we have reinforced animal behavioral core with the ARRA supplementary fund. Last time, we reported that we had completed behavioral assays of nNOS knockout mice and the manuscript has been accepted and published. As backcross breeding of PCM1 knockout mice has been completed, we are doing behavioral assays. Finally, longitudinal behavioral characterization of mice with genetic modulation via in utero RNAi transfer is done as the first level to address the role for DISC1 interactome (including nNOS, PCM1, and other interactors) in adolescence. This project has ended and all funds were fully exhausted. |
