This application proposes the purchase of a new bone histomorphometry system (OsteoMeasure XP Advanced System) consisting of a digital video analysis system and a fluorescence microscope. This equipment will allow the PI of this application, Dr. Bellido, and personnel in her laboratory to increase the speed in the analysis of experiments proposed under the parent grant entitled “Osteocyte control of bone formation via Sost”. The scope of the project is the same as the one formulated in the parent grant. The overall goal of the project is to advance understanding of the control of bone formation by osteocytes and to elucidate the contribution of these cells to the osteoblastogenic actions of PTH, PTHrP, and mechanical stimuli. The hypothesis to be tested is that activation of PTHR1 signaling in osteocytes leads to a rapid and direct inhibition of Sost expression, which, in turn, is responsible for increased bone formation in response to systemic elevation of PTH as well as to bone loading through local increase in PTHrP.

Quarterly Activities/Project Description This award supported the purchase of a new bone histomorphometry system (OsteoMeasure XP Advanced System) consisting of a digital video analysis system and a fluorescence microscope and additional small instruments. This equipment allowed the PI of this application, Dr. Bellido, and personnel in her laboratory to increase the speed in the analysis of experiments proposed under the parent grant entitled “Osteocyte control of bone formation via Sost”. We continue to use the equipment purchased with this award in a very active way. Several abstracts were presented reporting the work performed, one study was published, another study was submitted for publication and is being reviewed after a revision, and three other manuscripts reporting work being completed are under preparation. This has been indicated in the final progress report already submitted to the NIH. Work is complete and this is the final report
