At 95% complete, equipment is in place & technology is integrated. Adjusting to loss of key staff with approaches to ensure technical expertise is maintained & instruments & knowledge base are sustainable. Focus on research & increasing impact of instruments through utilization by students, staff, & faculty & broadening use to new project areas, with emphasis on sustainability. All instruments comprising the immuno/cell biology cluster are used in support of MRI research goals. New policies ensuring security, training, & responsible use were developed this quarter to be instituted next quarter. Project 3 Cramer/Dolan: A mouse trial involving intranasal vaccination of a model antigen (OVA) & plant-made interleukin 12 was completed. The goal was to assess immune baselines for strong cell-mediated response comparing vaccination in the presence of different adjuvants. Spleens & regional lymph nodes were collected & used to establish primary cultures for analysis by flow cytrometry. The Miltenyi Biotec gentleMACS Dissociator was critical for processing samples providing excellent homogeneity & viability. Analyses demonstrated IL-12 mediated shift in immunity toward CMI with plant-derived cytokine & will support a publication on in vivo efficacy of plant-made product. Upgrades to BD Pathway 855 Bioimaging System enabled development of powerful new assay for assessing enzyme delivery & disease phenotype correction in a cell model of human lysosomal storage disease (Acosta & Cramer). Fibroblasts (normal, Hurler syndrome, & Hurler/Scheie syndrome) were labeled with DAPI (labels nuclei to define ?one cell?) & with Lysotracker-red to stain lysosomes. Demonstrated that lysosomal area per cell was significantly different between normal & diseased fibroblasts & enzyme-mediated lysosomal ?correction? of phenotype could be effectively quantified. The assay provides significant advantages in statistical measurement & efficiency (replaces a 4 week protocol with a 4 day protocol). |
