The proposed program will lead to efficient bioconversion of carbon dioxide into an infrastructure-compatible liquid biofuel, n-butanol, without the requirements of photosynthesis. We will accomplish this through three major innovations supported by ARPA-E: genetic modifications of bacteria that assimilate carbon dioxide, oxygen, and hydrogen in the dark; development of an industrially scalable bioreactor system for sustainable production of butanol with these organisms; and a novel approach to recovery of the butanol from the bioreactor. Hydrogen bacteria normally use carbon dioxide, oxygen, and hydrogen for growth, but we will short-circuit the normal cellular pathways so that the organisms will convert these gaseous inputs into butanol. Several levels of genetic modifications will enhance the efficiency of carbon dioxide bioconversion, so that the organisms can support an industrial-grade process. This process will be a significant improvement over the current state-of-the art for biologically-produced butanol, because we anticipate at least a twofold improvement over current levels of productivity in a cost-effective manner, so that the product will outcompete ethanol with respect to price, and be comparable to gasoline. Furthermore, an innovative approach will be taken for generating the gaseous substrates, using a proprietary technology that converts waste biomass into carbon dioxide and hydrogen.

For the eleventh quarter of this project, progress was achieved on each of the major objectives. In some cases, alternative and better strategies were developed to achieve the goals. These are discussed. The following accomplishments were achieved and address expected milestones:
-all mutant and knockout and knockin strains constructed thus far for this project grow with doubling times on CO2 well within the 12 – 24 hour doubling time of the listed milestone for this quarter. In addition, progress was made on butanol optimization and strain construction.
-To examine whether existing strains could be scaled up for butanol production, it was found that GMO#1 produced trace butanol with gluconate in the disk stack reactor
-GMO#1 did not produce butanol under autotrophic conditions in the disk stack reactor
-Both gas stripping and pervaporation processes were further tested and optimized.
-Process models for gas stripping are being developed and validated with experimental data being collected.
