Despite the advent of new “targeted” drugs for the treatment of blood cancer "leukemia"), many patients will fail therapy and relapse. The goals of this proposal are to develop and test new leukemia therapies, based on transfusion of cells from the blood and immune system, which are designed to eliminate small numbers of cancer cells that persist in drug treated patient. These “cellular therapies” should provide import new treatment choices that increase the chance of permanent cure for people living with leukemia.

Quarterly Activities/Project Description In the past Quarter, progress has been made on Specific Aim 1 of the proposal. Aim 1 (To determine the mechanisms of the GvL effect of adoptive T-cell and NK-cell immunotherapy of BCR-ABL–induced CML-like leukemia in mice). We have induced CML-like disease in TNFRI/II dko cells (from Dr. Warren Shlomchik, Yale); in preliminary experiments the disease appears to be fully susceptible to allogeneic DLI, suggesting that TNFalpha is not required for GvL. We have obtained humanized CD19 mice from Dr. Tom Tedder at Duke, and will assess the utility of huCD19 CAR-modified NK cell immunotherapy against BCR-ABL1-induced B-ALL. These donor mice are now out of quarantine and are being bred. We are intercrossing two specific strains: huCD19 transgenic mice, and Cd19–/– knockout mice. The initial cross has generated progeny that should be Cd19+/– heterozygotes, and 50% will carry the huCD19 transgene. These progeny are being phenotyped by FACS analysis of PBL using anti-CD19 mAbs specific to human or mouse CD19. Trangenic progeny will then be backcrossed Subsequently, we will generate BCR-ABL1-induced B-ALL in this genetic background, and assess the in vitro sensitivity of these leukemic blasts to anti-hu-CD19 CAR-modified NK-92 cells and primary murine NK cells. The work is complete and this is the Final Report.

Description of Jobs Created In work supported by this ARRA-funded grant, we used resources to support the employment of a postdoctoral fellow, Dr. Cristina Gavrilescu, in the Van Etten laboratory. Without this support, Dr. Gavrilescu’s position would have been eliminated. During the 2.5 years of support from this grant (currently in no-cost extension), Dr. Gavrilescu was able to increase her exposure to new techniques in molecular biology/oncology, including proficiency at generating and titering retroviral vectors, performing BM transduction and transplantation, analyzing leukemic mice, and carrying out therapeutic testing of molecularly targeted drugs delivered by oral gavage or parenteral injection. She participated directly in the experiments carried out under this application and is a co-author on a manuscript recently published in Blood (Distinct graft-vs-leukemic stem cell effects of early or delayed donor leukocyte infusions in a mouse chronic myeloid leukemia model. Lu YF, Gavrilescu LC, Betancur M, Lazarides K, Klingemann H, Van Etten RA. Blood. 2012;119:273-284). In December 2011, Dr. Gavrilescu left the Van Etten laboratory to accept a position as a senior scientist at a biotechnology company in Cambridge, MA. We believe she represents an excellent example of the impact of ARRA/NIH funding in promoting job creation in the health sciences sector in the United States.
