The proposed longitudinal study is designed to examine reading development at critical points in its establishment (from ages 7.5-10.5) in nonimpaired (Nl) and reading disabled (RD) cohorts. Our previous cross sectional research has identified reading group differences in both functional neuroanatomical and behavioral trajectories; the proposed longitudinal study is aimed at gaining a better understanding of behavioral, neurobiological, and genetic etiological factors responsible for this observed divergence. We examine the hypothesis that the candidate etiological agent that might underlie variation in neurodevelopmental and behavioral trajectories is gamma-aminobutyric acid (GABA): research has shown that GABA plays a critical role in learning and memory. Accordingly, the proposed research will permit relating developmental changes in reading performance and functional neuroanatomy for reading (measured with fMRI) to GABA expression, measured with magnetic resonance spectroscopy (MRS) and genetic analyses, linking polymorphisms in the GABA family genes to GABA expression in the brain. Specifically, the research aims to: 1) Characterize concurrent relations among genetics, neurochemistry, functional neuroanatomy, and reading behavior at 2 important timepoints in reading development, 2) Investigate Nl and RD differences for each of these measures, 3) Examine subsequent developmental trajectories in Nl and RD cohorts, 4) Better characterize learning capacities and learning styles in these cohorts, 5) Develop dynamic brain/behavior phenotypes sensitive to genetic analyses, and 6) Contrast multi-level profiles and developmental trajectories in subgroups of RD.

Quarterly Activities/Project Description The ARRA grant yoked with the R01 entitled ?Neurobiological Foundations of Reading and Learning in Reading Disability? (Kenneth Pugh, PI) is a one-year project examining MRI and fMRI data from a longitudinal tracking study designed to examine reading development at three critical points (Time 1 = 7.5 years, Time 2 = 8.5 years, and Time 3 = 9.5 years) in non-impaired (NI) and reading disabled (RD) individuals. The ARRA funded project adds a new focus on intensive analysis of structural MR data acquired during fMRI scanning sessions at Time 1 and Time 3. To work on this we hired a Post-doctoral Associate (Dr. Peter Molfese), under supervision of Dr. Hemant Tagare and Dr. Ken Pugh, is examining structural MRI data and linking it with both fMRI and cognitive measures to identify those brain/behavior patterns that discriminate NI and RD cohorts at different time points. To date, automatic volumetric calculations for 362 participants have been performed using Freesurfer 5.1. Freesurfer allows for cortical parcellation based on gyral and sulcal structures, or the calculation of volume within structures of the brain. Data were next exported to the SAS statistical software for analyses. Individuals who had brain regions with volumes identified as statistical outliers (greater than 2 standard deviations above the mean), were rechecked. At this point, enough data has been processed from both the first and third time points to begin longitudinal analyses of anatomical brain data. To date, statistical analyses have begun to investigate how changes in behavioral measures collected at the first and third time point corresponding to changes in anatomical brain structure volume and cortical thickness. We have also begun preparing data for final analyses using subgroups within our sample comparing children with speech sound disorders and children who developed the ability to speak later than typically developing children. This is the final quarterly report.
