"This award was for a 3-year Director's opportunity award entitled ""Translating Dosage Compensation to Trisomy"". Our lab has long studied inactivation of one X-chromosome which provides for equal dosage of genes between males and females. While basic science aspects of X-inactivation are widely studied as an important model for developmental control of gene regulation, the Lawrence lab is also interested in the clinical relevance of this for understanding genetic disease. Having the proper dosage of genes can be as important as having the correct sequence of genes, thus understanding the dosage compensation mechanism of the X-chromosome can have relevance to human conditions in which chromosomal ""dosage"" is abnormal. The most common and clinically important example of this is trisomy 21 which causes Down Syndrome. Currently there is little therapeutic option for individuals who carry defects of chromosomal dosage, and our goal is to extend insights from normal human X-chromosome inactivation in directions that provide better understanding of chromosomal dosage disorders and may eventually lead to therapies. Since X-chromosome dosage compensation occurs in very early embryonic cells, this project focuses on studying abnormalities of chromosomal dosage in human and mouse iPS cells. This approach uses skin or other somatic cells that are induced to have properties of pluripotent stem cells, avoiding the ethical issues raised by use of human embryonic stem cells."

Quarterly Activities/Project Description "As promised in the last quarterly report, we submitted a second round of revision (by invitation) for a manuscript that will show we accomplished the top priority of this project, submitted to one of the top biomedical journals. This manuscript demonstrates success at the central ""high risk, high innovation"" goal of this project, and represents a major advance in translating understanding of dosage compensation for the X chromosome to problems of abnormal dosage compensation, such as in Down Syndrome. Work for revision was completed and the manuscript resubmitted one month ago, so we hope to hear soon. We are also making progress on extending this project to the mouse model system, and have generated key reagents for this purpose. However, we needed to repeat the targeting work because the first transgenesis was not successful, for technical reasons related to transgene expression as well as accuracy of targeting. We are constructing new transgenes designed for two different mouse models."
