Quarterly Activities/Project Description This project has ended as of 8/31/2012. Antigen coding sequences (pre-membrane and envelope) of all 4 dengue serotypes were optimized for human codon usage, and the optimized DNA sequences were synthesized and cloned into plasmid vectors. Using these clones, two recombinant adenovirus vectors each expressing antigens from two dengue serotypes were constructed. This work was done with Genvec, the consortium partner, using its proprietary adenovirus vector containing modified hexon gene. The two bivalent vaccine constructs were characterized by DNA sequencing and antigen expression in infected 293 cells by indirect immunofluorescence and western blot assays. A murine immunogenicity study was completed in which the vaccine constructs were demonstrated to elicit neutralizing antibody response in vaccinated mice. Vector seed stocks were prepared which will be useful in future cGMP manufacturing of vaccine constructs. Because these constructs were based on new, modified adenovirus vectors, we have also completed the necessary process development and analytical development to aid in cGMP manufacture of vaccines. Sufficient material (GLP reagent grade) was produced for confirming immunogenicity in monkeys. A non-human primate study (funded separately) was started in October 2012. The constructs were immunogenic in rhesus monkeys; neutralizing antibodies to all 4 serotypes were demonstrated in animals vaccinated with the tetravalent (mixture of two bivalent vaccines) dengue vaccine. This project started 16 months after the initial funding, necessitated by a change in the consortium partner and revision to statement of work, all of which was articulated to and approved by NIH. Project has made significant progress since then; however, cGMP manufacture of the vaccine could not be completed because a no-cost-extension to the grant was not granted.
