Twin studies have provided insights into multiple disease and developmental endpoints. Because their genetic and environmental similarities are easily understood, twins can elucidate the contributions of genetic and environmental factors to illness and health. Twin studies have also integrated molecular methods to examine gene-environment interactions and epigenetics. Although many countries claim to have registries, only those in Scandinavia are large population-based twin registries. The few American twin registries that exist have shortcomings that hamper the advancement of research in the US. The two registries that approximate the population from which they were drawn, the WWII Veteran and Vietnam Era Twin Registries, have only male twins restricted by age and lack of racial and ethnic diversity. The state-based registries, for example the Mid-Atlantic and Minnesota Twin Registries, have been constructed by identifying twins from birth records which are then linked to driver license information. However, these registries disproportionately miss female twins who have married and taken on their husband's surname. They also include participants who may not be representative of the reference population. The University of Washington Twin Registry (UWTR) was established to study the genetic and non-genetic factors that affect health. The Registry is an ongoing collection of twins identified from the State of Washington Department of Licensing. Unique to Washington, the Department of Licensing asks every applicant if s/he is a twin to avoid issuing duplicate license identification numbers to twins. After obtaining relevant permissions, we began collecting the necessary information to construct the Registry. Currently, ~3,000 twin pairs are enrolled in the Registry and 280 twin pairs have participated in the sample and data repository. This application is to significantly expand the UWTR in order to fully establish this unique resource and accelerate the pace of research in understanding and treating multiple disease and developmental endpoints.

"Start-up Activities": Washington State Vital Statistics data linking is underway and should be completed shortly. "Milestone 1: Twin Recruitment": Twin enrollment at the end of the grant period is 7,205 pairs. With the infrastructure made possible by this award, recruitment will continue and we expect to enroll 100-200 pairs per month for the foreseeable future. Collection of DNA sample kits and phenotyping surveys for "Milestone 2: Twin Repository" was very successful. The repository has 8,868 DNA samples and 8,736 phenotyping questionnaires with a pairwise rate of approximately 80% for a total of 3,561 pairs with both DNA and survey data. For "Milestone 3: Environment & Health" we are continuing to geocode all new UWTR twins when they enroll in the Registry with the set of built and social environment exposure measures we have developed."Milestone 4: Multilevel Pain Study" has completed the in-person data gathering phase (200 pairs complete). Final sample processing is in progress and preliminary data analyses are starting.In terms of "Milestone 5: Data Sharing/Collaborations," 1 R01 will be reviewed on October 5, 1 R01 (>$500,000 direct costs) and 1 R21 are in preparation for October submission, with 3 R01 submissions in preparation for February 2013. Several manuscripts are under review and in preparation including a description of the Registry for a Special Issue of Twin Research and Human Genetics (submitted on September 17th). |
