SANFORD-BURNHAM MEDICAL RESEARCH INSTITUTE


The overall goal of the project is to characterize the structures and interactions of the BCL-2 family proteins, the principal regulators of the mitochondrion-dependent pathway for programmed cell death or apoptosis. Their opposing pro- or anti-apoptotic activities are exerted through a network of interactions with partners within and outside the BCL-2 family, both in the cytosol and in intracellular membranes. Their associations with each other and with mitochondrial membranes have particularly important consequences for mitochondrial physiology and for cell fate. Their functions are intimately related to a large number of major human diseases (cancer, neurodegeneration, AIDS, etc.). This Supplement supports studies that are currently in progress and that are expected to yield significant results in the next few months. It provides additional funded time and resources for a senior postdoc who is working on the project, and funds to purchase new laboratory equipment.

This Supplement supported studies that are currently in progress and that are expected to yield significant results in the next few months. It provided additional funded time and resources for a senior postdoc who is working on the project, and funds to purchase new laboratory equipment. Support from this supplement has enabled us to complete and recently publish one important study regarding cell death protein-protein interactions. The work is published in the journal Biochemistry (Yong et al., 2009, Biochemistry 48:8704-8711). Recently, we have also been able to purify and obtain structral data for pro-apoptotic BAX, a protein that had been difficult to prepare. Studies on the interactions with its partners are ongoing. THe supplement has enabled us to initiate a collaboration with Don Newmeyer (La Jolla Institute for Allergy and Immunology) to examine binding of inhibitor small molecules to BAX. The additional results obtained with support of this supplement have allowed us to submit a revised renewal application for the parent grant R01GM065374 for the March 2010 deadline.

Description of Jobs Created This supplement provided additional funded time and resources for one senior postdoc currently working on the project. This postdoc has been working on the project since 2006. He is highly skilled in NMR spectroscopy and biochemistry, and has extensive experience with the structural and biochemical aspects of the BCL-2 proteins. First-author publications describing his work on this project have been submitted or and one paper has been published since this supplement was funded. Without additional support this position would have had to be terminated, and he would have been forced to pursue an alternative career. The Supplement has provided additional funded time to develop the project and foster a new promising career in scinece and technology.
