This award is designed to investigate the role of protein kinase C epsilon (PKCe) in the antiadrenergic action of adenosine. The antiadrenergic action of adenosine via adenosine A1 receptors limits the responsiveness of the heart to beta-adrenergic nerve stimulation in the normal, ischemic and aging heart. PKCe is important in the signal transduction of adenosine A1 receptors which are activated by myocardial adenosine. The information garnered from this project will lead to the development of cardiac agents and therapies that should help to alleviate heart failure.

Project Activities Description Heart Diseases Research
Quarterly Activities/Project Description Final experiments have been conducted investigating the effects of adenosine and adrenergic agent stimulation of mammalian ventricular cardiomyocytes on the activity and translocation of PKC epsilon. In our investigation of the effect of adenosine on p42 MAPK and the involvement of PKC epsilon, it became apparent that adrenergic stimulation was in some manner affecting the primary stimulation. These studies are revealing a complex interaction in the heart between adenosine and catecholamine stimulation at the level of PKC epsilon. Final data reduction is being conducted and manuscript preparation is being initiated to report these findings. In last quarter of this project Western blotting techniques were performed to comfirm our earlier findings. A manuscript has been perpared and is ready for submission dealing with his important findings. The final invoicing for the project will be complete in the next couple weeks.
