The MGH/MIT Morris Udall Center of Excellence in PD Research is taking a broad, collaborative and interactive approach to the study of Parkinson's disease. The Projects address critical questions concerning the selective vulnerability of dopamine neurons, the mechanism and consequences of Lewy body formation and alpha-synuclein aggregation, the neural systems consequences of parkinsonism and synuclein pathology, and molecular approaches for modifying this pathology. These issues will be explored using a range of systems, from yeast genetics, to mammalian cell culture, to rodent models to human postmortem material. The Center incorporates state-of-the-art technologies including high throughput yeast genetic screens to identify modifiers of synuclein aggregation and toxicity, viral vector gene transfer to study factors in mammalian cell culture and rodent models, multi-unit tetrode recordings to study striatal plasticity, fluorescence lifetime imaging to study protein-protein interactions, and laser capture microdissection and gene arrays to study transcriptional dysregulation. The Center has a Clinical and Training Core that provides care to patients with Parkinson's disease, gathers data on clinical features of the disease and response to therapy, solicits brain donations for neuropathological study, and trains outstanding clinician scientists to be future leaders in the field. The Center also has a Bioinformatics Core that serves to integrate and analyze data across the projects, and facilitate sharing of the information. The Administrative Core is charged with management of the Center and facilitating the sharing of information, ideas, and reagents among the investigators and with other components of the Udall Centers consortium. The investigators of the MGH/MIT Center are dedicated to a program of collaborative and interactive studies which will lead to better treatments for people with Parkinson's disease.

Quarterly Activities/Project Description 2) Quarterly Activities/Project Description The overall purpose of the Clinical and Neuropathological Core is to provide support the research mission of the MGH/MIT Udall PD Center through clinical and neuropathological activities. The clinical results will be measured by the establishment of the Parkinson Longitudinal Cohort, a cohort of men and women with PD and related parkinsonian disorders along with controls who participate in our ongoing clinical research studies into the cause, prevention and cure of PD. To date at the end of fourth quarter, the Core has enrolled in the Parkinson Longitudinal Cohort 34 normal control subjects, 71 Parkinson's Disease, 15 Parkinson's Disease with Dementia, and 23 Dementia with Lewy Bodies cases. The neuropathological results of the Core will be measured the autopsies and neuropathological examinations performed. To date at the end of fourth quarter, the Core has performed eight autopsies: two Parkinson?s disease cases and six Dementia with Lewy Bodies cases.

Description of Jobs Created 1) The MGH/MIT Udall Parkinson Disease Center was not renewed on the first round of our competitive renewal but has a no cost extension to 8/31/2010. We intend to submit a revised application in December, 2009. We are concerned that in this interim funding lapse several personnel will have to be let go. This Administrative Supplement award will allow us to collaborate to keep them here. The projects and cores will pursue the specific aims of the original proposal that was funded previously. The Center has provided important contributions to our understanding of Parkinson?s disease. Dr. Hyman has developed assays of alpha-synuclein misfolding and aggregation and is now examining ways to disrupt these conformational changes. Dr. Lindquist has identified modifiers of alpha-synuclein toxicity in yeast and is exploring the mechanism of the toxic process. Dr. Graybiel has shown that parkinsonian rodents do not learn normally and their striatal activity doesn?t go through plastic changes during the acquisition of a behavior. In collaboration with Project 4, she identified a correlation of CalDAG-GEF1 and 2 proteins with the development of levodopa-induced dyskinesias. TRH is similarly regulated. Project 4 has also discovered gender-specific alterations in gene and protein expression in the substantia nigra and the temporal cortex of control, PD and DLB brains. The Administrative Core supports all the projects and cores. The Neuropathology/clinical Core, Core B, supports the longitudinal cohort, Projects 1 and 4. The Bioinformatics Core supports projects 2, 3 and 4. This award is specifically an Administrative Supplement to Core B of the MGH/MIT Udall PD Center. The goal of Core B is to support the research mission of the MGH/MIT Udall PD Center through clinical and neuropathological activities. To this end, we have established and will follow a cohort of men and women with PD and related parkinsonian disorders who participate in our ongoing clinical research studies into the cause, prevention and cure of PD. This cohort ? the Parkinson Longitudinal Cohort ? will undergo annual neurological and neuropsychological examinations to characterize the motor and cognitive aspects of PD; they will donate blood samples for genetic and biomarker studies. We will follow these individuals over time, obtain autopsies at death and perform detailed neuropathological examinations to correlate with the prospectively collected clinical, behavioral and biomarker data. Brain tissue from these subjects will directly support research projects of the Udall Center conducted by Drs. Young (Project # 4) and Hyman (Project # 1) as well as investigators at other institutions. Clinical Core activities will also have an impact beyond our Udall Center through our training of young investigators in caring for patients with PD, supplying well-characterized patients to externally-funded clinical research projects, and characterizing the clinical and biomarkers features of PD over time in light of neuropathological findings. We did submit a new Udall Center application in December, 2009.
