In the most recent data supplied by the American Heart Assocation, cardiovascular disease remained the major underlying cause of death in the United States with the majority of these deaths being due to coronary heart disease and stroke. Given the high prevalence of atherothrombotic disease and high MI and death rates, and incidence of adverse effects (bleeding and other safety issues), there remains a high unmet need for new therapeutics as exemplified by PZ-128, that can target both collagen and thrombin-dependent activation of platelets without unduly affecting hemostasis.

GLP safety pharmacology respiratory studies in guinea pigs and non-GLP monkey safety cardiology studies have been completed by Lovelace under the SMARTT program. Under the new P50 HL110789, Phase I Drug product dose validation, stability, and sterility studies have been initiated and IND assembly is continuing as reports are received from MPI, and SRI. The work is complete and this is the Final Report.

Dr. Ping Zhang's position (Postdoctoral Fellow) was retained as a result of this award. Dr. Zhang is conducting cyno monkey platelet aggregometry and biocompatibility studies, in addition to writing reports for the upcoming Dec IND submission to the FDA. Dr. Zhang is a platelet expert and expert in arterial thrombosis models--two rare skill sets that are essential to the success of the project. Dr. Lidija Covic is a long-standing collaborator with extensive platelet and pepducin experience. Dr. Covic is responsible for training and overseeing human platelet experiments and animal model efficacy studies. She lends her expertise in use and administration of pepducins in the work outlined in the original proposal. Principal Investigator; Athan Kuliopulos. |
