This program will continue to study fundamental structural properties of the cell nucleus that support biological control and are disrupted in leukemia and metastatic breast cancer. We will gain insight into components of nuclear organization that can be targeted for innovative cancer therapies.This Administrative Supplement for the parent P01 Nuclear Structure and Gene Expression? requests funds to expedite and enhance key collaborative objectives among Program Project investigators. We continue to pursue our hypothesis that parameters of nuclear architecture support cell growth and phenotypic properties of normal and tumor cells by facilitating the organization of chromosomes, genes, transcripts and regulatory complexes as dynamic three-dimensional micro-environments within the nucleus. We propose the following Supplemental Approaches to continue to characterize functional relationships between nuclear organization and fidelity of gene regulation: Supplemental Approach 1: Characterization of chromatin-mediated mechanisms supporting tumorigenesis-related gene regulation at subnuclear domains. We will perform high resolution chromatin mapping of Runx controlled genes and Runx dependent post-translational modifications of histone proteins that require fidelity of subnuclear targeting in leukemia and breast cancer cells, as well as during three-dimensional breast epithelial tissue development in culture (collaboration between Projects 1, 2 and 4). Supplemental Approach 2: Examination of spatio-temporal and epigenetic parameters of subnuclear targeting and chromosomal association of Runx proteins during interphase and mitosis. We will apply high resolution immunofluorescence microscopy including time-lapse analysis of live cells and biochemical analyses to define temporal-spatial and functional relationships between Runx and Brg1 proteins and selected Runx-related co-factors. We will also define epigenetic states of Runx foci during interphase and mitosis in normal and cancer cells (collaboration between Projects 1, 2 and 4). This Supplement will enhance our capabilities to execute the original Specific Aims of the P01 Program and will expedite the discovery of epigenetic mechanisms linked to subnuclear targeting that are deregulated in cancer.

High-resolution chromatin analysis (ChIP-DeepSeq) of leukemic cells has been finalized and a manuscript describing these results is near submission. ChIP-DeepSeq analyses of breast cancer cells using multiple antibodies, as well as Brg1-mediated regulation of growth of breast cancer cells, will continue as part of the parent P01 grant. Immunofluorescence analyses of co-regulatory proteins will be performed as necessary to support other studies in the parent P01. The final expenses will be invoiced in the next couple weeks |
