Quarterly Activities/Project Description ▲ The completion of toxicity and cell culture based potency assays of selected high potency compounds will be pursued in the next quarter. Aim 1.1. First part, virtual screening for identification of DENV protease inhibitors by Dr. David Ostrov has been completed and the leads will be validated in the next quarter (Padmanabhan and Ostrov) ▲ At the Wichita State University supported by sub award, progress related to (a) the optimization of two novel series of compounds based on the 1, 2-benzisothiazolinone and isoindolin-1-one scaffolds led to submission of the following manuscript already published (Groutas and Padmanabhan). ▲ Several manuscripts are in either preparation or has been submitted for publication and are at different stages of review process as detailed below. ● Srinivasan, P., Boonyasuppaya, S., Saperstein, S., Nagarajan, K., and Padmanabhan, R. Evaluation of compounds of 8-hydroxyquinoline scaffold as inhibitors dengue virus protease by in vitro and cell-based replication assays (in preparation) (Padmanabhan?s lab). ● A manuscript entitled, ?Inhibitors of Dengue Virus and West Nile Virus Proteases Based on the Aminobenzamide Scaffold by Sridhar Aravapalli, a Huiguo Lai, b Tadahisa Teramoto, b Kevin R. Alliston, a Gerald H. Lushington, c Eron Ferguson, a R. Padmanabhan, b William C. Groutas a* submitted early in the last quarter was peer-reviewed, revised and resubmitted in this quarter and is now accepted and published in Bioorg. Med. Chem 20, 4140-4148, 2012. ● A new manuscript has been submitted for publication entitled, ?Design, Synthesis and Characterization of Novel 1,2-Benzisothiazol-3(2H)-one and 1,3,4-Oxadiazole Hybrid Derivatives: Potent Inhibitors of Dengue and West Nile Virus Proteases?
