Project Title IMMUNOLOGIC UNIQUENESS OF THE FEMALE GENITAL TRACT IN HIV PATHOGENESIS

Quarterly Activities/Project Description Project 1 To determine the role of vaginal DCs in HIV-1 entry and dissemination, we established protocols to isolate human vaginal mononuclear cells and construct vaginal explants by adapting our protocols for intestinal cell isolation and explant construction. Project 2 In vitro studies on the effect of medroxyprogesterone acetate (MPA) based contraceptives on immune processes. The effect of MPA, progesterone, and estrogen on the ability of T cells and monocytes/macrophages to produce relevant cytokine responses following stimulation was studied. MPA exerts a highly significant immunosuppressive effect on these cell populations and inhibits production of IFN-gamma, TNF-alpha, and other important cytokines and chemokines following stimulation. Project 3 We demonstrated that MPG suppresses the production of IFN- and TNF- by purified mucosal mononuclear cells isolated from the vaginal and endocervical mucosa of premenopausal and postmenopausal women. In a separate set of studies we showed that MPG inhibits HIV-1-specific T cell responses as determined by ELISPOT and IFN--production assays to a significantly higher degree than progesterone alone. Core B During the first year of this application we have enrolled 15 women and 2 males used as controls the majority of whom were African American (76.5%). From these women we have collected blood, menstrual blood, and vaginal fluid samples. These samples were processed in our Center for AIDS Research sponsored processing and storage facility. Furthermore, the majority of these samples were used by the Mestecky and Hel laboratories to accomplish the aims set forth in projects 2 and 3 of our program. Also obtained were vaginal, uterine, and cervical surgical samples that were used to accomplish the goals of project 1. Any remaining funds may be deobligated.
