Tanycytes are specialized cells that line the floor and ventrolateral walls of the third ventricle. Compelling evidence suggests they have an important role in generating thyroid hormone due to high expressionof type 2 iodothyronine deiodinase, an enzyment critical for conversion of the prohormone, T4, into the biological active thyroid hormone, T3. We have proposed, therefore, that tanycytes are involved in th regulation of the hypothalamic-pituitary-thyroid (HPT) axis and energy homoestasis. To facilitate the study of these cells in neuroendocrine regulation, we will identify tanycyte specific genes by laser capture microdissection and microarray analysis to develop transgenic mice that express Cre recombinase specifically in tanycytes.

Because of concerns that our initial RNA-Seq analysis of tanycytes removed by LCM from the wall of the third ventricle may have also contained other cell types that are adjacent to or within the wall of the third ventricle, including arcuate nucleus AGRP/NPY neurons, infiltrating inflammatory cells (macrophages) and ependymal cells, we decided to repeat the RNA-Seq analysis on tanycytes using a different strategy that would yield highly purified material. As nestin is highly and selectively expressed in tanycytes in the adult animal, we obtained Nestin-GFP transgenic mice and purified the tanycyte population by FACS. After several attempts, we developed an approach that allowed extraction of high quality material for RNA-Seq analysis. This analysis is currently being conducted and should be available within the next two week. The resulting transcriptome will be compared to our initial data set to determine whether tanycytes can be characterized as multipotent cells with properties of inflammatory cells, stem cells and hormonal cells, as indicated in the initial RNA-Seq analysis. We are also in the process of verifying that Rax is selectively expressed by tanycytes in the adult brain using in situ hybridization histochemistry. We have identified RAX as another gene that demonstrates high and selective expression in the initial data set of our RNA-Seq analysis. Once confirmed (as expected), we will move forward to prepare a transgenic mouse in which Cre-recombinase is selectively produced in Rax-expressing cells under the regulation of tamoxifen.
Results of this work were presented before the annual meeting of The Endocrine Society in June, 2011, and a second paper has been submitted for presentation before annual meeting of The Endocrine Society to be held in June, 2012. Once we have confirmation of the tanycyte transcriptome in our most recent RNA-Seq analysis, the data will be prepared for publication. The work is complete & this is Final Report.
