During the next two years this project will test the hypothesis that abnormal imprinting of the IGF2 gene is a biomarker for colorectal cancer (CRC) predisposition, which exerts its increase risk via the IGF axis, resulting in specific pathological and molecular characteristics and a unique CRC phenotype. During the next two years of this award the following specific aims will be pursue: 1) To determine whether LOI of IGF2 in lymphocytes is associated with proliferation index, apoptosis index, and micro-vascular density of colorectal cancer; 2) To determine whether LOI of IGF2 in peripheral lymphocytes is associated with rectal cancer recurrence in individuals with Duke?s B/C rectal cancer.

Quarterly Activities/Project Description We have completed recruitment of colorectal patients as per our sample size calculation. There are three manuscripts in preparation including 1) Association of LOI of IGF2 with non colorectal cancer 2) Evaluation of IGF2 mRNA, tissue IGF2, and serum IGF2 with abnormal imprinting of IGF2 3) Risk of recurrence of colorectal cancer among IGF2 deficient patients. We have completed our first no-cost extension and requested a second no-cost extension to complete analysis by John Hopkins.
