SEATTLE UNIVERSITY
Seattle University (SU) has received an award from the NSF MRI-R2 program to purchase a large-volume preparative ultracentrifuge to support programs in undergraduate research and education. The large-volume preparative ultracentrifuge is initially being used by three different faculty members and undergraduate student researchers to conduct basic research projects. Dr. Stenbak and her students use the ultracentrifuge to investigate the molecular requirements for assembly of foamy viruses (ancient, non-pathogenic retroviruses). Cell fractionation through ultracentrifugation allows students working with Dr. Smith (microbiology/biochemistry) to isolate and characterize bioactive compounds involved in territoriality from Myxococcus sp. Finally, students working with Dr. Murphy (biological chemistry/molecular pharmacology) use the centrifuge to prepare eukaryotic and prokaryotic cell lysates for characterization of Hsp90-based molecular chaperone protein function and activity.

The overall goal of this award was to purchase and install a large-volume ultracentrifuge at Seattle University to support faculty research and research training efforts. The ultracentrifuge was successfully acquired and installed and is being used in laboratory classes as well as faculty research. Specific activity in Autumn 2012 involved continued harvesting of virus particles to support undergraduate research projects studying Foamy Virus assembly in the Stenbak lab. During this quarter, continued progress was made analyzing a panel of deletion mutants for defects in viral protein processing and virus assembly. The preliminary results found three different regions within the Foamy virus polymerase protein that appear to be required for either viral cleavage, virus assembly, or both. These mutants are undergoing further characterization in a cell-based bifluorescence assay for protein-protein interaction defects to support the initial findings. Two undergraduate research students participated in this project during the fall quarter and our results were presented at the M.J. Murdock Charitable Trust Conference for Undergraduate Research. These studies have made significant progress and the use of the ultracentrifuge purchased with this grant was, and will continue to be, critical for the success and publication of this work. Equally as important, 6 undergraduate research students and over 30 undergraduate science students had the opportunity to work with and learn from this instrument. Although the grant period is ending, it is clear that students and faculty will be able to utilize this ultracentrifuge for many years to come and this will continue to greatly enhance our research and educational goals at Seattle University. The cost of the equipment was less than the total award amount; therefore the ?Total Federal Amount of ARRA Expenditure? field does not equal the amount in the ?Award Amount? field. No more funds will be expended. |
