Inhibition of androgen synthesis is the current treatment of human diseases like prostate cancer and polycystic ovary syndrome. This approach is suboptimal. By defining how the enzyme responsible synthesizes these androgens, we hope to establish new approaches to treatment. Our study will apply to other enzymes that metabolize drugs.

Quarterly Activities/Project Description The final Total Federal Amount of ARRA Expenditure for this project is less than the Award Amount. The project was completed under budget. No more funds will be expended on this project The purposes of this award are: 1) to determine if eliminating cytochrome b5 from the mouse testis by homologous recombination using Cre/loxP technology significantly impairs testosterone synthesis and causes undervirilization of male mice; 2) to determine of the same residues on cytochrome b5 are required for stimulation of various cytochrome P450 activities. Regarding Aim 1, we have repeated the enzyme assay with testicular homogenates, and we have confirmed the defective 17,20-lyase activity in the cytochrome b5 testis knockout animals. We have begun analyses of serum steroids, and while basal testosterone appears normal, the 17-hydroxyprogesterone is markedly elevated. We are planning stimulation experiments and urine steroid analyses to expand these results. Thus, Aim 1 continues to progress well, in part because we have now established the LC-MS/MS assays for the serum steroids, and we are very pleased with the assay?s performance characteristics. I have relocated the laboratory to the University of Michigan, and we have now resumed P450 enzyme expression and assay experiments for Aim 2. I have hired a qualified scientist with specific expertise in cytochrome P450 work for this project. In addition, with Paul Hollenberg in Pharmacology, who is an expert in CYP2E1, we will be better positioned to studies these P450 enzymes other than CYP17A1 with our panel of cytochrome b5 mutations. We have received a no-cost extension, and the grant has been closed out at UT Southwestern in preparation for transfer to my new institution.
