| Quarterly Activities/Project Description |
• The development of a novel influenza vaccine approach is continuing at the VPPL with support from the VCMP. This approach, identified as HA-ferritin nanoparticle vaccine (HA-ferritin), involves the engineering of fusion proteins expressing influenza hemagglutinin with a self-assembling H. pylori ferritin nanoparticle. VCMP activities supporting this project include procurement of four polyclonal and monoclonal antibody test reagent preparations for the HA-Ferritin nanoparticle project. These reagents will be used to develop assays for process development and potentially GMP production. In addition to a previous clone showing positive binding to HA-ferritin A/New Caledonia, clones have been confirmed with reactivities to the naked ferritin nanoparticleHA-ferritin A/California and HA-ferritin A/Singapore. A recent decision by the VRC may lead to the requirement to develop specific reagents for A/Indonesia, an H5 type.
• The VCMP successfully received IBC registration to allow E. coli-based process development in laboratories at the VPP. No studied have been planned to-date as preliminary work at the VRC has shown the E. coli approach to render improperly folded nanoparticles.
• The VCMP successfully established a new project through a task order with Pfenex, Inc. to investigate the use of a proprietary Pseudomonas expression system for bacterial expression of nanopartilces. This project is in early stages.
• The VCMP is continuing the stability evaluation for products previously produced under this project.

The positions include Quality Control Analysts, Development Associate, and Manufacturing Associate. The QC Analysts are responsible for drafting, executing, and reviewing qualifications of analytical instrumentation; calibrating and maintaining lab equipment; applying knowledge of cGMPs and GLPs; participating in out-of-specification and failure investigations; and maintaining the safety of the lab. The Development Associate III will assist in developing scalable processes with improved product yield and reduced costs for manufacturing systems. Additionally, assist with maintenance of production equipment. Works on complex problems where analysis of situations or data requires an in-depth evaluation of various factors. May determine methods and procedures on new assignments and may provide guidance to other lower level personnel. The Manufacturing Associate performs in-lab activities and manufacturing. |
