Infection of children with Shiga toxin (Stx) producing Escherichia coli (STEC), primarily a food and waterborne aquired disease, is the leading cause of hemolytic-uremic syndrom (HUS) in the US. There is no specific treatment to prevent or ameliorate HUS. We propose to develop aptamer-based therapeutic agents for prevention or treatment of HUS.

Quarterly Activities/Project Description We generated ssDNA aptamer pool by employing one of the most common methods, asymmetric PCR amplification which produces ssDNA due to the unequal concentration of sense and antisense primers in the reaction mixture. In the past few months, we optimized assymetric PCR by using different ratios of sense and antisense primers (like 1:20, 1:40, 1:80, 1:160) to generate ssDNA efficiently. In addition, we also performed 3 rounds of ssDNA selections with N23 pool using nitrocellulose membrane filtration method. The filter binding assay to estimate the enrichment of the pool with ssDNA and RNA aptamers binding to Stx2 is undergoing.

While the work of the postdoc should be guided by the senior investigator, the person in this position is expected to contribute ideas and suggestions to the research and to publish results. The individual in this position may be given other guided responsibilities such as supervision of staff, grant proposal writing, writing papers, etc. to prepare for an independent scientific career.
