UNIVERSITY OF TEXAS SOUTHWESTERN MEDICAL CENTER AT DALLAS
During the past quarter, we have made excellent progress in establishing Dr. Anwarul Ferdous as an independent investigator in the Division of Cardiology here at UT Southwestern Medical Center. He is swiftly establishing an independent program of research on the biology of FoxO transcription factors in cardiovascular development and disease. I n addition, he is moving forward with a research program on microRNAs during cardiovascular development. He has hired a new research assistant and purchased the requisite equipment and supplies needed to launch his independent career. All of this is being supported either by the P30 or by institutional resources. The original objectives as laid out in the P30 application are unchanged, and we are presently on pace according to the timelines that we originally proposed.

No more funds will be expended.Project#1a:Transcriptional regulation of cardiovascular systems by FoxO1.We completed our initial goal to understand the role of FoxO1 in development.We demonstrated that FoxO1 governs Vcam1 expression, and that FoxO1-Vacam1 axis is intimately involved in cardiovascular as well as placental morphogenesis(Ferdous et al, 2011 and Ferdous and Hill 2012).To define the independent role of FoxO1 in cardiovascular development,we utilized lineage tracing study,which suggests that FoxO1 is involved in valve development/function. Project#1b:Transcriptional regulation of cardiac remodeling by FoxO1.he Ferdous lab is also utilizing a conditional FoxO1 knockout mouse model for myocyte-specific ablation of FoxO1 gene and to investigate its role in cardiac remodeling following pressure overload.We identified that inactivation of FoxO1 blunts pathological hypertrophy.We identified that the FoxO1-thyroid axis participates in pathological remodeling of heart following stress.Project#2:Transcriptional regulation of myogenic differentiation&cardiac fibrosis by Ets-family transcription factors.The genesis of vascular and muscular progenitor cells from a common ancestor is known, yet transcriptional regulation of this alternative cell fate remains elusive.We found that Fli1 acts as a critical node of vascular vs. muscular fate of the progenitor cells.Project#3:Effects of posttranscriptional regulation of connexin 43 expression by micro-RNA on cardiac function and remodeling. Using luciferase plasmid harboring the 3? untranslated region of Cx-43,miR-19a expression plasmid and virus,we demonstrated that forced expression of mir-19a attenuates Cx-43 expression in vitro and in vivo & that attenuation of Cx43 levels affects calcium homeostasis in neonatal rat cardiomyocytes.Project#4:Identification&characterization of ubiquitinated proteins in autophagic cardiomyocytes. |
