Quarterly Activities/Project Description We published our second to last article in Biochemistry 2011, 50:9788-9796. The title is "The HIV-1 monomer induces a specific interaction with LTR DNA for concerted integration". We have made significant progress on purifying HIV-1 integrase monomer to ~1.9 mg/ml. This data and others allow us to successfully obtain a R21 NIH grant entitled "HIV-1 Integrase Structural Biology" with a Impact Score of 21 in March, 2012. We have submitted a article entitled ""The Asymmetric C-terminal Domain Dimer of Rous Sarcoma Virus Integrase Serves as a Viral DNA Binding Surface" for publication. These studies were done in collaboration with Dr. H. Aihara, University of Minnesota.Here we report the crystallograph characherization of a 3-domain integrase protein fully capable of concerted integration as well as a high-resolution crystal structure of a 2 domain integrase fragment.
