HIV causes AIDS in humans. Integration of HIV DNA by integrase into chromosomes is necessary for virus replication which cause AIDS. We investigate the mechanisms associated with integration and prevention of integration by clinical inhibitors.

We published our second to last article in Biochemistry 2011, 50:9788-9796. The title is "The HIV-1 monomer induces a specific interaction with LTR DNA for concerted integration". We have made significant progress on purifying HIV-1 integrase monomer to ~1.9 mg/ml. This data and others allow us to successfully obtain a R21 NIH grant entitled "HIV-1 Integrase Structural Biology" with a Impact Score of 21 in March, 2012.
We have submitted a article entitled ""The Asymmetric C-terminal Domain Dimer of Rous Sarcoma Virus Integrase Serves as a Viral DNA Binding Surface" for publication. These studies were done in collaboration with Dr. H. Aihara, University of Minnesota.Here we report the crystallograph characherization of a 3-domain integrase protein fully capable of concerted integration as well as a high-resolution crystal structure of a 2 domain integrase fragment. |
