Quarterly Activities/Project Description Glioblastoma multiforme (GBM): We had previously developed a stable cell line for the signal transducer and activator of transcription 3 (STAT3) luciferase reporter. However after several cell passages, luciferase expression decreased and the STAT3 reporter specificity was lost. This forced us go back and work with a transient reporter system in which the reporter plasmid is transiently transfected into cells and expression measured 48 hours after plasmid transfection. We have optimized the transient transfection conditions, implemented a multiplexed assay for the simultaneous measurement of the STAT3 activity and cell viability. Using this assay we validated the assay with commercially available positive control compounds and then screened 250 compounds previously shown to bind STAT3 through Broad's small molecule array (SMM). We have identifies 31 compounds which decreased STAT3 activity by 20% of control (untreated) level; three of these compounds decrease STAT3 activity by at least 30% of control. We are further characterizing these effects by fine tuned titration studies. Additionally, Dr. Jose Silva is investigating whether the SMM identified STAT3-specific binding compounds reduce STAT3 phosphorylation. T cell acute lymphoblastic leukemia (T-ALL): Based on reviewer comments to a manuscript submitted to Nature, we are currently investigating the mechanism of reversal of glucocorticoid resistance via AKT inhibition in primary patient samples.We are reporting a status of fully completed this quarter and did not utilize all the funds.
