The proposed studies are focused upon identifying and inhibiting the pathogenic process by which a high-fat or western-style, diet evokes Type 2 diabetes, using the mouse model of this common mammalian syndrome. Preliminary work completed supports the hypothesis that pancreatic beta cell failure to transport glucose is a fundamental pathogenic trigger that can elicit metabolic defects including insulin resistance, and may be further resolved to a coordinate defect in protein glycosylation linked to the glucose sensing mechanism of the pancreas. This research aims to provide a molecular and cellular understanding of hog the high-fat diet contributes to the pathogenesis of Type 2 diabetes - a current global epidemic in the Human population, and seeks to establish a novel approach by which disease pathogenesis may be averted.

Quarterly Activities/Project Description This research will contribute to establishing the physiologic roles of mammalian glycans, using the laboratory mouse as a model system and with a focus among the glycan linkages that promote hybrid and complex N-glycan branching. Findings from this project may reflect the key role of pancreatic beta cell glucose transport in suppressing disease pathogenesis, as implied by our preliminary data among mice bearing similarly enforced expression of Glut-2. Specific Aim 1: To establish and characterize offspring of multiple transgenic mouse founder lines bearing enforced expression of the GnT-4a glycosyltransferase specifically in pancreatic ß cells, and test for resistance to pancreatic ß cell failure and the pathogenesis of Type 2 diabetes in response to a high-fat diet. Aim 1 has been completed and the results have been published in the journal Nature Medicine (Ohtsubo et al., 2011) defining a pathogenic pathway leading from diet to obesity-associated diabetes. Discoveries reported identify the origin and molecular mechanisms underlying the current human epidemic in obesity-associated Type 2 diabetes. Our findings suggest a more rational approach that involves identifying resistance alleles among obese individuals that do not acquire Type 2 diabetes. Our discoveries further reveal new approaches by which the pathogenesis and severity of Type 2 diabetes may be averted. Specific Aim 2: Test the role of the Glut-2 glucose transporter as the GnT-4a substrate responsible for conferring resistance to ß cell failure and the pathogenesis of Type 2 diabetes in response to the high-fat diet. For Aim 2, a significant and relatively complex breeding remains in progress, and these studies could be completed following funding the R01-A1 renewal application, which is currently in review.

Description of Jobs Created Jobs created and filled/positions retained: This project requires a postdoctural fellow to be responsible for breeding and phenotypic characterization of GLUT2 transgenic mice as well as different mouse strains that exhibit distince responses to the high-fat diet, including analyses of glucose stimulated insulin secretion, Glut-2 expression, Mgat4a expression, and protein glycosylation by GnT4a. A graduate student will assist phenotyping and characterizing of mouse strains susceptible and resistant high-fat diet-induced Type 2 diabetes. This position will further work closely with other lab members to investigate mechanisms of transcriptional regulation of Mgat4a and Glut genes, and begin to characterize glycoprotein and lectin-dependent interactions modulated in the beta cell by Mgat4a function. Jobs retained: It is necessary for a more senior postdoctoral fellow to be responsible for MGAT4A transgenic mouse studies to characterize physiological outcomes, and to investigate the transcriptional regulation of Mgat4a and Glut2. The principal investigator will be responsible for overall direction and supervision regarding the research projects; several meetings per week will be held with the post-graduate and graduate student researchers to review data and prioritize experimental plans. The PI will also engage directly in research experiments and administrative duties that support the projects.
