REGENTS OF THE UNIVERSITY OF MICHIGAN


This award is funded under the American Recovery and Reinvestment Act of 2009 (Public Law 111-5). This Major Research Instrumentation-Recovery and Reinvestment (MRI-R2) award funds the development of two versatile, modular fluorescence microscopy platforms for tracking and mechanically probing single molecules/particles at the University of Michigan (UM), and the installation of a third, user-friendly microscope system for user training and project development. The development and application of two open-access single molecule tracking and probing microscopes transforms the limited capability for high-content screening of biological samples. Benchmarks are set that make both instruments operational within two years and lay out plans for testing, further optimization and expansion over an additional two years, after which recharges will maintain them. The instruments are being utilized for research and education by the University of Michigan community and beyond, engaging physical and biological scientists as well as an engineer. This development activity helps establish a unique, nation-wide, educational Center of Single Molecule Analysis (CoSiMA) in a highly cross-disciplinary, emerging field of transformative power in the biosciences. CoSiMA is engaging in vigorous outreach efforts to experimentalists, theorists, undergraduates, and high school students with large, local, underrepresented groups, as well as the public at large, in close collaboration with UM's existing IDEA Institute. These efforts are facilitated by the creation of a central laboratory facility with access to multiple single molecule microscopes under guidance of a Steering Committee and a senior staff scientist, as well as by the broad inspirational appeal of the emerging single molecule biosciences. Results from the studies enabled by the new equipment are disseminated by student and faculty presentations at regional and national meetings, and through publication in peer-reviewed journals.

Project Activities Description Chemistry
Quarterly Activities/Project Description We are upgrading the existing confocal microscope to perform pulsed interleaved excitation (PIE). The supercontinuum laser will be spectrally split into two wavelengths using a dichroic. The longer wavelength will be delayed by 25 ns and the laser is pulsed at a frequency of 20 MHz (50 ns between pulses), which will result in alternating excitation pulses. Optics and hardware required for this are being purchased and assembled. Two graduate students from Biophysics, one undergraduate student from Chemistry and a postdoctoral fellow from the Medical school were added to the users’ roster. The graduate students and the undergraduate are independent, the postdoc’s experiments are performed by SMART center staff. We have purchased an astigmatic lens and a lens holder/switch to implement 3-dimensional imaging on the Single particle tracker. Axial localization is adjudicated based on the intensity profile of the sample under investigation – particles in focus have evenly distributed radial intensity profiles whereas particles just above or below the focal plane have elliptical/ellipsoidal intensity profiles elongated along different major axes. Two new users were added to the roster; both are postdoctoral fellows from the Medical school. One user is independent, while the other postdoc’s experiments are performed by SMART center staff. An Environmental Cell was purchased for the TT-AFM to scan samples in liquids/buffer. Previously, samples were scanned in air, a non-native environment for biological macromolecules. Two new users from Chemistry were trained on the AFM by SMART center staff. An additional (third) postdoc was hired at 50% to complement the existing 100% and 50% postdocs and complete the assembly of the Single Particle Nanomanipulator. Final optical, hardware and software parts of the assembly are currently being purchased. Final assembly and initial calibration of the system are expected to begin in early April.
